Michail Antonio opened the scoring with a header on 28 minutes before Robert Snodgrass netted the Irons' second just three minutes later. Lucas Pérez added a third to ensure a dominant lead before half-time.
Ryan Fredericks powered in the pick of the goals on 51 minutes and Angelo Ogbonna put the home side five goals ahead with a free header just minutes after.
Snodgrass got his second, and West Ham's sixth, on 60 minutes, before debutant Grady Diangana scored twice, combining with fellow Academy of Football graduate Joe Powell for each goal to wrap up the 8-0 win.
Victory over Macclesfield Town was West Ham United's biggest win in any competition since beating Bury 10-0 in October 1983.
